Chemiluminescence Determination of D-amino Acid Based on Immobilized Enzymes

Abstract: D-amino acid oxidase and HRPwere separately immobilized on chitosan adsorbed on the silica gels to develop a chemiluminescent method,which was combined with two oxidation reactions of D-amino acid producing H2O2 catalyzed by D-amino acid oxidase and of Luminol-H2O2 generating CLcatalyzed by HRPtogether,also was stopped──flow technique used in this system. It has a good selectivity and sensitivity. Under optimized conditions,the detection limit is 1.8 ×10-5g/mL,the liner range of D-amino acid concentration was from 8× 10-8 to 1. 4×10-6g/mL,It was practicable to determine D-amino acid in serum directly with satisfactory results.
