毛细管电泳柱后化学发光检测装置的研制

Construction of End-Column Chemiluminescence Detection System for Capillary Electrophoresis

摘要: 报道自行组装的毛细管电泳柱后化学发光检测装置。鲁米诺和N-4-氨基丁基-N-乙基异鲁米诺-H2O2体系用于评价该装置的可行性,得到发光试剂鲁米诺和ABEI较尖锐对称的电泳分离峰,检出限分别为50和290fmol;基于一些金属离子对鲁米带-H2O2的快速动力学催化反应,得到Cr(Ⅲ)和Zn(Ⅱ)离子非常尖锐的分离峰,理论塔板数达1.51×105和1.86×105,检出限为15amol和2fmol.

Abstract: Aself-constructed end-column chemiluminescence detection system for capillary electrophoresis has been described.Luminol and N-4-aminobutyl-N-ethylisoluminol(ABEI)H2O2 chemiluminescence systems are used to illustrate the feasibility of this instrument.Comparatively sharp and symmetric electrophoretic peaks are obtained with the detection limit of 50 fmol and 290 fmol for luminol and ABEI, respectively, Cr(Ⅲ) and Zn(Ⅱ) ions are separated with rather sharp peaks due to their fast catalytic effect on the luminol-H2O2 reaction.The theoretical plate number reaches 1.51×105 and 1.86 ×105 for Cr(Ⅲ)and Zn(Ⅱ),the detection limits of which are 15 amol and 2.07 fmol,respectively.
