Abstract: Linear and supercoiled DNAwere cleaved by HPLCpurified seryl-histidine dipeptide(SH). It was found that the DNAfragments produced by the reaction of SHand DNAcould be ligated together by T4 DNAligase. This result implied that the SHwas the first example of the ion free artificial DNAcleavage agent that could split DNAby hydrolysis mechanism.

Key words: Seryl-histidine dipeptide, DNA, Cleavage, Hydrolysis mechanism
