Studies on the Separation and Extraction of α-Amylase and Proteinase from Fermented Liquor of α-Amylase by Aqueous Two-phase Partitioning

Abstract: Aqueous two-phase system composed of PEG/(NH4)2SO4 was once to isolate α-amylase and proteinase from fermented liquor of α-amylase in this paper. The optimum extraction conditions were: 15% PEG1000, 20% (NH4)2SO4, pH=8, the partition coefficient of α-amylase was as high as 19.6 with a yield of 90%. The separation coefficients between α-amylase and proteinase were 15.1 and 1.5 folds increase of specific activity compared with those the original fermented liquor. The yield of proteinase in water phase was more than 60%. The preliminary study on back-extraction condition was done in this paper.
