Voltammeric Determination of DNA with Doxorubicin Hydrochlorldum as Intercalator on Graphite Electrode

Abstract: Asynthesized 75-mer single stranded DNA (ssDNA-1) was covalently immobilized on a oxidized graphite electrode surface that had been activated using EDCand NHS.The immobilized ssDNA-1 hybridized with various concentrations of complementary ssDNA-2 in hybridization buffer containing DXH.The dsDNA/DXHsystem was formed on the electrode surface.The cathodic waves of the DXHbound to the dsDNAwith cyclic voltammetry were used for the determination of ssDNA-2.The reduction peak currents of DXHwere linearly related to the concentration of ssDNA-2 between 1×10-5 to 5×10-7 mg/mL.The determination demonstrated the method is practicable.
