Determination of Complement Level in Serum by Polarographic Immunoassay

Abstract: Anew method for determination of complement level in serum by polarographicimmunoassay was developed in this paper.This method is based on the reaction between ophenylenediamine and hydrogen peroxide catalyzed by hemoglobin from the sensitized cells incomplement-mediate hemolysis, producing an electroactive compound 2,2'-diaminoazobezen(DAA)and determining the DAAby linear-sweep polarography.The linear calibrationcurve is obtained in the range of 0.10 to 1.00 CH50 unit/mLand the limit of detection is 0.08 CH50 unit/mL.The sensitivity of the method is ten times more than that in the spectrophotometric method reported.And 25 samples at a time can be determined.The proposedmethod has been applied to the determination of complement level in healthy human serumand patient serum.
