Capillary Electrophoresis Coupled with Tris(2,2'-bipyridyl) Ruthenium Electrochemiluminescence Detection for Bupivacaine Determination

Abstract:

The determination of bupivacaine by capillary electrophoresis with Ru(bpy)32+ electroche-miluminescence detection was described.The detection potential,Ru(bpy)32+ concentration,phosphate buffer concentration and pH,injection voltage and time were investigated.Experiment results showed that a good separation for the bupivacaine in plasma can be achieved under the optimized conditions,such as 10 s at 10 kV of electrokinetic injection,10 mmol/L separation phosphate buffer(pH=8.0),15 kV of separation voltage,5 mmol/L Ru(bpy)32+,and 50 mmol/L phosphate buffer(pH 6.5) in the ECL cell.The linear range was 0.02-10μg/mL,the relative standard derivation(RSD) was 3.4%,and the detection limit was 3 ng/mL.When the method was applied to measure bupivacaine in rat plasma,the results were satisfactory.
