The Effect of the Concentration of the Buffer in the Capillary Zone Electrophoresis with PEG-400 Cross Linked Capillary Column

Abstract: The effect of concentration of the buffer on the migration time,column efficiency,selectivity,resolution and the sampling amount were investigated in CZEwith the PEG-400 cross linked capillary column.Agroup of the peptides were used as the sample.The elution order of about 60% substance pairs changed when the concentration of the buffer changed only from 0.01 mol/Lto 0.04 mol/L.Such effect has been used as the foundation for the optimization of the condition.The peak height and area were increased with the increase of the same amounts of the sample.The relationship between the peak height and the concentration of the buffer was approximately linear.Attention should be paid when one calculates for the sample amount with the expression to the electric injection mode,where the concentration in the sample pool is different from that in the column.
