NMR自扩散研究脂肪酸与α-环糊精的相互作用

摘要: 用脉冲梯度场NMR测量自扩散系数的方法研究了乙酸、丙酸和丁酸与α-环糊精在低于临界浓度条件下的相互作用.研究发现,3种脂肪酸的扩散系数均随着环糊精含量增加而减小,其减小趋势与烷基的大小有关,烷基越大,其扩散系数减小越快.利用测得的扩散系数计算出3种脂肪酸和环糊精形成复合物的平衡常数,发现平衡常数随烷基链增长而增大,该结果与2D-ROESY(RotatingFrameOverhauserEffectSpec-troscopy)谱均表明,在脂肪酸与α-环糊精相互作用过程中,烷基与环糊精内腔的疏水相互作用起主导作用,不同脂肪酸和α-环糊精的结合比均为1:1.

Abstract: The association between α-cyclodextrin and three kinds of short chain fatty acids was studied by NMR self-diffusion measurements. Dilute solution condition was adopted to avoid precipitating of the solutes. The diffusion coefficient of fatty acid in the solution, which reflects the strength of association between α-cyclodextrin and fatty acids, was found to decrease with increasing alkyl chain length of fatty acid. Together with 2D-ROESY experiment, therefore, it was demonstrated that in forming inclusion complex, the hydrophobic interaction is predominant among various possible interactions between α-cyclodextrin and fatty acids.
