Cholesterol Biosensors Based on Cholesterol Oxidase Immobilized in a Silica Sol-gel Matrix on a Prussian Blue Modified Electrode

Abstract: Acholesterol biosensor prepared by immobilization of cholesterol oxidase in a silica sol-gel matrix on the surface of Prussian Blue-modified glass carbon electrode was reported. The properties of the sensor were investigated. It is based on the detection of hydrogen peroxide produced by ChOx at 0. 0-0. 1 V. The measurements conditions were optimized. Cholesterol can be determined in the concentration range of 5 × 10-7~8 × 10-5mol/Lby voltammetry or 5 ×10-6~5 × 10-4mol/Lby amperometry. The method gives a high sensitivity owing to the electrocatalytic layer of Prussian Blue to the electro-reduction of enzymatic production of hydrogen peroxide. In addition, the effect of easily oxidizable substances presented in samples such as ascorbic acid and uric acid can be minimized.
