Preparation and Characterization of Protein A Immobilized on Monolithic Capillary Column for Affinity Chromatography

Abstract: An assay technique for the determination of the human IgG in human plasma has been developed by utilizing protein A immobilized monolithic capillary column. The affinity chromatography experiment was performed on a capillary electrophoresis instrument by using its pressure system as the driving force. Reactive monolithic capillary columns have been prepared by in-situ copolymerization of the monomers in the presence of porogenic dilute, and protein A was immobilized on the monolithic rods directly or through a 11 carbon atom space arm with molded synthetic methods. The non-specific adsorption of two kinds of media has been studied and the results showed that the medium without space arm gives very low non-specific adsorption of BSA. The affinity column without space arm was used to determine the human immunoglobulin G(HIgG) in human serum. The correlative coefficient of the calibration curve reaches 0.998 7. The total time of rapid analysis was less than 0.7 min. The consumption of eluent buffer and sample is much low than by conventional HPLC.
