Abstract: The optically active(2R,8R)-bis(hydroxymethyl)-1,5,9-triazabicyclo[4.4.0]dec-9-ene has been synthesized from D-asparaginate with a total yield of 25.4%.In the synthesis, the key intermediate is a thiourea derivative from reduction and coupling of the starting materials and the key reaction is it's methylation and double SNreaction to form bicyclic skeleton.Amino and hydroxy are protected by tosyl and silyl groups which can be cleaved under very mild conditions respectively. The target molecule is a useful intermediate in the construction of chiral and selective receptors of organic anions.
