Studies on the New Packings of High Performance Ion Exchange Chromatography for the Separation of Proteins(Ⅱ)─The Synthesis and Characteristics of the New Packings or Weak Cation Exchange Chromatography

Abstract: The studies on the synthesis and chromatographic characteristics of three kinds of new packings of high performance weak cation exchange chromatography for the separation of proteins have been reported on the basis of previous work.The silica reacted with γ-glycidoxypropyltrimethoxy silane, then the product reacted with polyethyleneimine and yielded a crosslinked product, which reacted with a cycloanhydride to give WCX-5 and WCX-6 packings;If the crosslinked product reacted with polysuccinimide, followed by hydrolysis, the WCX-7 packings have been prepared.The crosslinkage of packings ligand could raise the recovery of the bioactivity of proteins, The recovery of the bioactivity of lysozyme is over ninety four percents.The column capacity of WCX-7 packings is bigger than that of other two packings because the crosslinked ligand of WCX-7 is more than that of other two. The reten tion rule of some proteins on the WCX-7 column is different from that of the ordinary WCXcolumn.The retention of lysozyme and Cytochrome-c increases with the increase of pHvalue, The separation property of WCX-5 is the best.
