A Study of Abstraction of Hemoglobin by Non-organic Solvent Liquid-solid Extraction System

Abstract: Polymer-salt-aqueous liquid-solid extraction system is newly developed in 1990′s. The system has been successfully used for extraction of metal ions and bioactive substances such as malic dehydrogenase. The separation and extraction of hemoglobin by modified PEG 8000 mixed with Tween 80 and aqueous K3PO4 system is seldom reported. The easy phase formation by this method can make solid and liquid phases separated without any treatment, so that the imperfection of liquid biphasic technology can be eliminated. The method mentioned in this paper has the features of high capacity, cost effectiveness, convenience. Meanwhile the polymer and salt comprising the extraction system can stabilize and protect the bioactive substances. So the new technology is very suitable for the practical use in large scale. In the research, extraction efficiency from the solid phase is >99% and reversed extraction efficiency can reach 75%. The purity of hemoglobin is 99% after reversed extraction. Highly purified hemoglobin powder can be produced through dialysis and freeze drying treatment.
