Study of 1,2,4-Triazines—The Synthesis of 3-Methymercapto-5-Substituted Amino-1,2,4-Triazine-6-Carboxylic Acid Derivatives

摘要: 王序等报导3-甲硫基-5-羟基-6-甲基-1,2,4-三嗪(Ⅰ)在无水吡啶中与对-甲苯磺酰氯作用,倾入冰水中,得N4-[5′-(3′-甲硫基-6′-甲基)-1′,2′,4′-三嗪基]-3-甲硫基-5-氧代-6-甲基-4,5-二氢-1,2,4-三嗪(Ⅱ)[1.2]。(Ⅱ)在乙醇中与胺作用,得到3-甲硫基-5-取代氨基-6-甲基-1,2,4-三嗪及(Ⅰ)[3]。我们拟利用同样反应,自3-甲硫基-5-氧代-4,5-二氢-1,2,4-三嗪-6-羧酸乙酯(Ⅲb)[4]制备3-甲硫基-5-取代氨基-1,2,4-三嗪-6-羧酸衍生物,供抗肿瘤作用筛选。

Abstract: Ethyl-3-methylmercapto-5-hydroxy-1,2,4-triazine-6-carboxylate was treated in dry ice with Tscl to form ethyl N4-(5'-(3'-methylmercapto-6'-ethylcarboxylate)-1' ,2/,4/-triazinyl)-3-methylmercapto-5-oxo-4,5-dihydro-1 , 2,4-triazine-6-carboxy-late(Ⅳ).Compound (Ⅳ) reacted with a series of aliphatic and aromatic amines in etha-nol, and Ⅳa-b, Ⅶa-b, Ⅷa-c and Ⅸa-c were obtained respectively.
