Standardization for Retention Data of Drugs in RP-HPLC──A Study of Retention Index System

Abstract

Abstract: A retention index system based on alkan-2-ones was introduced for RP-HPLC.The effects of pH and methanol concentration in the mobile phase on RIwere examined.RI of seventy drugs and rnetabolites was determined using ODS-and phenyl-column with UVand RIDdetectors.The correlation between the lgk' and carbon number was linear from the second homologue.It has been proved that the RI decreases the variability of retention data and is suitable for the identification of drugs and the comparison of retention data obtained from different laboratories,instruments,columns,RIwas used for studying QSAR and QSRR of drugs and metabolites because there was a significant correlation between RIand lgPor π.
