Solid Phase Peptide Synthesis of Designed Yeast Transcriptional Activator Ac [Q236,Q242]GCN4(226─252)─GGC─NH2 and Its Dimerization

Abstract: Yeast transcriptional activator GCN4(226-252)-GGC-NH2 and the designed Ac-[Q236,Q242)]GCN4(226-252)-GGC-NH2 aiming at altering GCN4 DNA-binding specificity was prepared by stepwise solid phase peptide synthesis.It was found that the yield of the peptide would be greatly improved when treated with dithiothreitol(DTT).The dimer was obtained by air oxidation, which would be used in the sequence-specific DNAbinding activity experiments.
